چکیده

Background Mechanical stress is a primary determinant of bone growth and remodeling: weight bearing and locomotion stimulate interstitial fluid flow through the bone canalicular system, and the resultant shear stress is a major mechanism whereby mechanical forces stimulate bone growth [1]. In response to fluid shear stress and other types of mechanical stimulation, osteoblasts produce large amounts of NO, and genetic and pharmacologic studies indicate an important role of NO in osteoblast biology, but little is known about signaling downstream of NO in osteoblasts [1,2].

Src activation by cGMP/PKG II in osteoblasts: characterization of a mechano-sensitive signalling complex

Background Mechanical stimulation of bone cells is critical for maintaining bone mass and strength, and a better understanding of how mechanical stimuli are converted into intracellular signals to activate an anabolic program in osteoblasts/cytes is fundamental to improving treatments for osteo-degenerative diseases [1,2]. Weight bearing and locomotion stimulate interstitial fluid flow through ...

Non-Overlapping Functions for Pyk2 and FAK in Osteoblasts during Fluid Shear Stress-Induced Mechanotransduction

Mechanotransduction, the process by which cells convert external mechanical stimuli such as fluid shear stress (FSS) into biochemical changes, plays a critical role in maintenance of the skeleton. We have proposed that mechanical stimulation by FSS across the surfaces of bone cells results in formation of unique signaling complexes called mechanosomes that are launched from sites of adhesion wi...

The role of calcium channels in osteocyte function.

Osteocytic response to stretching, which is potentiated by PTH, is distinct from that of osteoblast to high frequency strain. A MAPK dependent signaling pathway is suggested in the osteoblast response. At least two different types of mechanotransduction pathways are present in bone cells of osteoblastic lineage.

Focal Adhesion Kinase Plays a Role in Osteoblast Mechanotransduction In Vitro but Does Not Affect Load-Induced Bone Formation In Vivo

A healthy skeleton relies on bone's ability to respond to external mechanical forces. The molecular mechanisms by which bone cells sense and convert mechanical stimuli into biochemical signals, a process known as mechanotransduction, are unclear. Focal adhesions play a critical role in cell survival, migration and sensing physical force. Focal adhesion kinase (FAK) is a non-receptor protein tyr...

Guanosine 3',5'-cyclic monophosphate (cGMP)/cGMP-dependent protein kinase induce interleukin-6 transcription in osteoblasts.

Natriuretic peptides and nitric oxide (NO) activate the cGMP/cGMP-dependent protein kinase (PKG) signaling pathway and play an important role in bone development and adult bone homeostasis. The cytokine IL-6 regulates bone turnover and osteoclast and osteoblast differentiation. We found that C-type natriuretic peptide and the NO donor Deta-NONOate induced IL-6 mRNA expression in primary human o...
